Application
E06 mAb-biotinylated has been used in immunodetection. It may be used in immunohistochemistry, western blot analysis and in in vivo imaging with magnetic resonance imaging (MRI) techniques.
The E06-Biotin is primarily used for sandwich ELISA determination of ox-LDL in serum or plasma samples. It can also be used in direct ELISA, competitive (IHC) and Western Blot analysis. For use in direct ELISA, make a 4 µg/mL working solution of 12 µg E06-Biotin in 3 mL of 1% BSA in PBS (1:62.5 dilution). Make a serial (1:1) dilution of this working solution to titrate the antibody. For competitive ELISA make a 2.5 µg/mL working solution (1:100 dilution).
Biochem/physiol Actions
The E06 antibody is a murine monoclonal IgM natural antibody that binds to oxidized phospholipid and reacts with oxidized Low-density lipoprotein (LDL), oxidized High-density lipoprotein (HDL) and protein covalently modified by oxidized phospholipid. This antibody has been used to probe atherosclerotic plaque in rabbits, monkeys, mice, zebrafish and humans. It also binds to apoptotic cells, but not viable cells.
General description
Avanti now provides high purity E06 from an animal-free culture of hybridoma cells. The E06 antibody was developed by Dr. Joseph Witztum at the University of California, San Diego using apolipoprotein E knock-out mice fed a high-fat diet for seven months.
